I think it's "Closer" but "Give Me Everything" could be a contender for his memorable hook.

At one point, I would have said So Sick or Closer, but now...I don't really think he has one? He obviously had many hits and a distinctive sound (well more Stargate's sound for a period, but he was integral to it too), but I don't think any of his individual singles have stuck around unfortunately. Even Irreplaceable and With You, as massive as they were in their day, have been superseded by other hits from the respective acts.
I'd actually agree with Give Me Everything, which isn't to discredit or belittle him. When a song has multiple hooks like that, it's a positive thing when yours is the one that most recall.

This is a bizarre case where I'd argue he doesn't really have one, in spite of several being big hits at the time. I know So Sick was the most successful back then, but I feel like it's all but forgotten today. Similarly, Closer and Miss Independent had a big impact at the time, but the longevity hasn't been there much either. Though more with Closer than the others I'd say. But Let Me Love You is probably what still has the most relevance now out of all of them. Yet it wasn't as big of a hit as some of his earlier ones. I guess I'll go with Closer, but I don't think there is one.

Because of You and SHE KNOWS (OMG!!!!) are my favorite singles from him as well. but So Sick was the first thing that popped into my head when I read signature song.

I legitimately can't imagine 99% of people under 20 having ever heard So Sick once in their life. Even I can barely remember it being big, and I would've been like 15.

I think I also like Give Me Everything as his signature hit, as a few others already suggested. His hook is what made it what it was.
On an aside, I still quite enjoy his album Year of the Gentleman. It's nothing groundbreaking, but it's a very competently made set of Pop-R&B tracks.
