Bjork, definately, but perhaps even moreso than her Kate Bush, whose unique style and genre-bending inspired Tori Amos, Bjork herself and pretty much all those alternative singer-songwriters. She's probably one of the most important women in 20th/21st century music.

Anyways, before even opening this thread, Bjork was the first that came to mind, definately.
And as for Missy Elliot, I agree, her sounds are very very unique, but are hard on my ears (those squeeky squeels of hers) and I can't distinguish one song from another. They use the same beats and sounds every time. Sorry any Missy fans out there, but this is how I feel.

Solange Knowles. "Feelin' You" was wildly original, but she isn't being given a far shot because of her sister. Her sound and voice are unique, she's very underrated, but I have no doubt she'll become a star in her own right one day soon.
